摘  要:本文根据化疗药物诱导肿瘤细胞产生凋亡的机制,采用细胞凋亡荧光染色法(AFS)分别检测了卵巢癌细胞株(3AO)、部分肺癌患者分离的癌细胞药物敏感性,并和传统的ATT法进行比较.用不同浓度的顺铂诱导3AO凋亡、两种方法学之间没有显著性差别(P>0.05);但用于肺癌患者分离的肿瘤细胞进行药物的筛选时,有显著性差别(P<O.05);AFS避免了MTT多种影响因素,比MTT快速、准确、敏感、重复性好,同时能观察凋亡、坏死及活细胞,有可能成为一种新的用于肿瘤化疗药物的筛选方法.Using in vitro established tumor cell lines as a target treated by the various concentrations cisplatium (DDP) and comparing with the conventionl MTT assay, we have set up an apoptosis fluorescence staining (AFS) in antitumor drugs sensitivity test. The results showed that there was no significant difiierence (P > 0.05) both in AFS assay and MTT assay to detect the apoptosis of the cell lines. However, there was significant difference ( P < 0.05) of the cytotoxic effect of the tumor cells derived from clinical samples. AFS assay is a rapid, accurate, reproductive and sensitive approach, which can observe three kinds of cell state i.e. apoptosis, necrosis and live cells at the same time. This method avoided the contaminated cells, cell fractions as well as the influence of other factors in MTT assay. It is possible to replace MTT assay for measuring cytotoxicity of antitumor drugs.
