摘　　要：目的探讨VEGF反义核酸对口腔鳞癌细胞的影响。方法利用脂质体将VEGF反义核酸导入口腔鳞癌KB细胞,采用ELISA法检测其培养上清液中的VEGF表达水平,用MTT检测肿瘤细胞生长状况,过氧化物酶的抗荧光素抗体法检测肿瘤细胞凋亡指数。结果经VEGF反义核酸处理后,肿瘤细胞的VEGF蛋白表达水平和细胞生长受到明显抑制,肿瘤细胞凋亡率显著增加。结论VEGF反义核酸通过阻止VEGF蛋白表达和促进细胞凋亡,对口腔鳞癌细胞有明显的抑制作用。[Objective] To explore the effect ofVEGF antisense nucleotide on oral squamous cell. [Methods] Antisense nucleotide was transfected into cells by liposome. VEGF expression was tested by ELISA. MTT assay were carried out to assess the proliferation potency. Apoptotic index was determined by means of anti-dioxyfluoran anti- body of peroxydase. [Results] After treated with antisense nucleotide, VEGF expression and growth of the tumor cells were inhibited obviously. Apoptotic rate increased. [Conclusions] Antisense nucleotide significantly suppress oral squamous cell through preventing VEGF expression and enhancing apoptosis.
