摘 要:目的粘附肽(精氨酸-甘氨酸-天冬氨酸)是细胞外基质高度保守的氨基酸序列,可与整合素家族受体特异识别并结合,诱导细胞附着。本研究应用人工合成的粘附肽片段,观察其对小鼠MC3T3-E1亚克隆14前成骨细胞的转谷氨酰胺酶表达的调节。方法MC3T3-E1亚克隆14前成骨细胞在常规培养和诱导分化条件下,分别加入粘附肽(GRGDS)和对照肽(GRADS),24h后用ELISA法测定细胞中转谷氨酰胺酶的活性。结果在诱导分化条件下,粘附肽(GRGDS)可以使MC3T3-E1亚克隆14前成骨细胞的转谷氨酰胺酶活性明显高于对照组和加入对照肽组(P<0.05)。结论粘附肽对小鼠成骨细胞分化早期细胞转谷氨酰胺酶的表达有促进作用。Objective To examine the effect of adhesive peptide on transglutaminase (TG) activity of osteoblast during early stage of differentiation. Methods MC3T3 - E1 ( subclone 14) pre - esteoblasts were treated with ascorbic acid to induce cell differemiation, and then GRGDS and GRADS peptide were added respectively. TG activity was assessed 24 h after the treatment by ELISA method. Results TG activity of GRGDS peptide group were increased significantly than the control group and GRADS peptide group ( P 〈 0. 05 ). Conclusion The GRGDS peptide could up - regulate TG activity of MC3T3 - E1/C14 esteoblast during the early stage of the differentiation.
