摘 要:目的研究不同浓度的17β-E2对鼠骨髓单个核细胞CD44、CD54表达的影响。方法通过免疫细胞化学及图像分析、统计学等方法来检测分析不同浓度的(10-6、10-7、10-8mol/L)17β-E2作用体外培养的SD大鼠骨髓单个核细胞不同时间(24、48、72h)后,细胞的CD44、CD54的表达。结果1超过其生理浓度的17β-E2使CD44的表达下降,17β-E2组随其作用时间的延长,CD44的表达呈下降趋势。217β-E2抑制骨髓单个核细胞CD54表达,其作用与剂量、时间的呈依赖关系。结论不同浓度的雌激素在不同的时间抑制了体外培养的骨髓单个核细胞CD44、CD54的表达,并可能由此影响细胞的分化。Objective To research the expression of CD44 and CD54 in marrow single-nucleus cells cultured with different concentrations of 17β-E2. Methods Immunocytochemistry, image analysis and statistic analysis were used to determine and analyse the expression of CD44 and CD54 in the marrow single-nucleus cells cultured with different concentrations of 17β-E2 and time. Results When the concentration of 17β-E2 was beyond physiological concentration, the expression of CD44 in the marrow single-nucleus cells was decreased. The expression of CD44 in all the marrow single-nucleus cells cultured with different concentrations of 17β-E2 was declined gradually as time went on. The expression of CD54 in marrow single-nucleus cells was inhibited by 17β-E2. The inhibited action of 17β-E2 was increased by the higher concentration of 17β-E2 or a longer time culture. Conclusion 17β-E2 can inhibit the expression of CD44 and CD54 in different concentrations and different time of culture, which is probably the channel for 17β-E2 to influence the differentiation of marrow single-nucleus cells.
