维生素E与维生素C抑制晶体上皮细胞凋亡研究

摘  要:目的探讨抗氧化剂维生素(VitE)、维生素(VitC)对H2O2氧化损伤所诱发的晶体上皮细胞(LECS)凋亡的抑制作用。方法在MEM培养基中离体培养兔晶体,分别在不同培养时限时观察对照组和各试验组晶体混浊情况,计算细胞凋亡率。结果H2O2+Vit E组、H2O2+Vit C组和H2O2+Vit E+Vit C组晶体混浊度都明显低于H2O2组晶体混浊度(P<0.01),说明它们能够抑制H2O2的氧化损伤作用,其中H2O2+Vit E+Vit C组的细胞凋亡率最高时才(11.22±0.25)%明显低于其他2组(P<0.01),抑制效果最好。结论抗氧化剂Vit E和Vit C可抑制H2O2氧化损伤所诱导的兔晶体上皮细胞凋亡及白内障的形成。Objective To investigate the inhibitory effect of vitamin E and vitamin C on the apoptosis of lens epithelial cells induced by H2O2. Methods The rabbit' s eye len were cultured in MEM medium, and the opacity of the rabbit's eye len was observed at the different time points of incubation, and the rate of the apoptosis was calculated. Results The opacity of the rabbit's eye lenses in H2O2 + Vit E group, H2O2 + Vit C group and H2O2 + Vit E + Vit C group was signifieantly lower than that of H2O2 group ( P 〈 0.01 ), which indicated these drugs could inhibit the apoptosis of eye lens epithelial induced by H2O2 The inhibitory effect was most obvious in H2O2 + Vit E + Vit C group, with apoptosis rate ( 1 1.22 ± 0.25 ) %, as compared with other groups ( P 〈 0. 01 ). Conclusion The antioxidants vitamin E and vitamin C could inhibit the apoptosisof lens epithelial cells induced by H2O2 and pathogenesis of cataract in rabbits.
