摘  要:目的观察脑缺血对AD大鼠皮层和海马异常磷酸化tau蛋白表达的影响,探讨脑缺血在AD进展中的作用。方法大鼠海马注射Aβ1-40成功建立AD大鼠模型后,于AD大鼠右侧纹状体注射内皮素-1建立临床常见的基底节区多发性腔隙性脑梗死的脑缺血模型,采用免疫组织化学法观察脑缺血对AD大鼠皮层和海马内Ser202位点异常磷酸化tau蛋白表达的影响。结果脑缺血后AD大鼠皮层和海马内异常磷酸化tau的表达较单纯的AD大鼠明显增多(P<0.05)。结论脑缺血促进AD大鼠皮层和海马内异常磷酸化tau的表达,进一步可促进神经纤维缠结的形成,提示脑缺血可促进AD的进展。Objective To investigate the role of cerebral ischemia in the progression of AD by observing the change of abnormally phosphorylated tau protein expression in cortical and hippocampal neurons of AD rats after cerebral ischemia. Methods AD rat model was established by hippocampal injection of Aβ1-40. Then AD rats were injected Endothelin-1 (ET-I) into the right striatum to establish cerebral ischemic model of multiple striatal lacunar infarction which is common clinically. Then immunohistochemical method was used to observe the effect of cerebral ishcemia on the expression of abnormally phosphorylated tau protein in cortical and hippocampal neurons of AD rats. Results Compared with the simple AD rats,the AD rats with cerebral ischemia showed significantly increased expression of abnormally phosphorylated tau in cortical and hippocampal neurons(P〈0.05 and P〈0.01). Conclusion Cerebral ischemia promote the expression of abnormally phosphorylated tau protein in cortical and hippocampal neurons of AD rats,which hints that cerebral ischemia can promote the formation of NFTs furthermore,and accelerate the progression of AD ultimately.
