脑室内注射对β-淀粉样蛋白大鼠海马BMP4和nestin表达的研究

摘　　要：目的通过侧脑室注射β-淀粉样蛋白(Aβ)后检测大鼠海马骨形成蛋白-4(BMP4)和巢蛋白(nestin)的表达,观察Aβ注射后局部神经干细胞(NSCs)的发生以及星型胶质细胞的活化增生情况。方法将Aβ1-42注射入大鼠侧脑室,分别于术后3、7、14、30d处死动物,应用免疫组化法观察BMP4的表达,用免疫荧光方法观察nestin,GFAP的表达,Brdu标记判断增殖细胞的种类。结果实验组的大鼠海马BMP4阳性细胞在3d已出现,到7d达到高峰,然后略微减少,维持在较高水平;而nestin阳性细胞也在3d已出现,到14d达到高峰,30d仍有表达,表达nestin的细胞主要为星形胶质细胞,部分活化星型胶质细胞来源于NSCs;对照组仅有极少数散在阳性细胞。结论β-淀粉样蛋白脑室注射后大鼠海马nestin表达增加,星型胶质细胞活化增生,BMP4可能促进了海马区NSCs向胶质细胞分化。Objective To observe the neurogenesis after unilateral i. c. v. injection of Aβ and the expression of BMP4 and nestin. Methods Adult male SD rats were divided into control group and experimental group randomly. Experimental group was infused with Aβ1-42 into the lateral ventricle and sacrificed 3d, 7d, 14d, 30d after infusion. The expression of BMP4 was observed by immunohistochemistry,while nestin,Brdu and GFAP by immunofluorescence. Results The BMP4 positive cells in the experimental group appeared in hippocampus on the 3d day and reached maximal expression on the 7th day after infusion of Aβ. The nestin positive cells also appeared on the 3d day and reached maximal expression on the 14th day. The nestin positive cells were mainly horizontal cell,and some of them origin from NSCs. They both decreased gradually after the peak. There were seldom BMP4 or nestin positive ceils in the control group. Conclusion The expression of nestin increases and astrocytes are activated after i. c. v. injection of Aβ,while BMP4 probablly promotes the NSCs in hippocampus differentiation towards astrocytes.
