摘  要:斑马鱼血管系统在原肠胚形成后不久便开始发育,血管系统的发育过程可分为血管发生和血管生成两个不同的阶段,其过程受到多种信号通路的调控,这些信号协同作用,以确保血管发育中的正常进行。本文主要以斑马鱼为模式生物,综述了血管发育遗传的最新研究进展及一些关键的信号调控通路,并介绍调节血管发育进程的一些关键的调控。以斑马鱼为模式生物来研究血管系统的发育遗传学,为理解人类血管的发育和再生,为缺血性疾病和肿瘤等疾病的治疗提供了新的途径。The vascular system develops shortly after gastrulation in zebrafish. The formation of the embryonic vasculature is initiated by the appearance of blood islands from progenitor cells in the visceral yolk sac. Within the blood islands, hemangioblasts differentiate into either hematopoetic or endothelial cells, with the former located within the channel space and the later cells lying on the edges. This first phase of blood vessel formation is known as vasculogenesis. A second phase then begins which is referred to as angiogenesis, involving active sprouting of vessels from pre-existing blood vessels. Interconnection of these primitive vessels results in the formation of a primary vascular plexus, which undergoes a complex process of remodeling. Eventually, the fetal vasculature emerges from angiogenic growth, selective fusion and regression of primitive ves- sels. In recent years, a number of different signaling pathways has been shown to be involved in vascular development. Key signaling pathways to vascular morphogenesis include vascular endothelial growth factor (VEGF), notch, etc. This review focuses on recent advances in embryonic vascular developmental genetics research, with special reference to the zebrafish. As the basic mechanisms and signaling pathways are well conserved between fish and mammals, the results of zebrafish studies will contribute greatly to understanding human vessel development. The disruption of vascular network can have devastating consequences; therefore, the study of vascular development will undoubtedly contribute to the improvement of therapeutics for vascular diseases ischemia diseases and cancer.
