摘 要:对固定化洋葱假胞菌G63脂肪酶催化大豆油和甲醇合成生物柴油的工艺进行了研究。通过甲基三甲氧基硅和四甲氧基硅缩水和水解反应形成的硅胶包埋固定化洋葱假胞菌G63脂肪酶。研究了酶用量、醇油摩尔比、反应温度、含水量对合成生物柴油的影响。优化的转酯化反应条件为:醇油摩尔比4:1,含水量5%,固定化脂肪酶15%,反应温度50℃,48h后转化率可以达到96.4%。固定化脂肪酶经多次使用后活力未见明显下降。Transesterification reaction was performed in non-aqueous conditions to prepare biodiesel using soybean oil and methanol as raw materials and immobilized lipase as catalyst. Lipase from Pseudomonas cepacia G63 was immobilized by entrapping with sol -gel prepared from tetramethoxysilane and methyltreimethoxysila. Using the immobilized lipase as catalyst, the effects of water content, enzyme dosage, ratio of methanol to oil and reaction temperature on the transesterification reaction were investigated. The results showed that the optimal conditions were as follows: 50 ℃, molar ratio of methanol to oil 4: 1,5% water and 15% lipase. Under the optimal conditions, methyl esters formation of 96.4% for reaction 48 h was obtained. The immobilized lipase proved to be stable and lost a little activity when was subjected to repeated uses.
