摘 要:目的探讨纳米载体对端粒酶反义寡核苷酸抑制肝脏肿瘤细胞生长的影响。方法制备壳聚糖纳米载体,并装载端粒酶反义寡核苷酸,用TRAP-ELASA法检测端粒酶反义核苷酸寡对肿瘤细胞端粒酶活性的影响,以MTT法测定端粒酶反义寡核苷酸对肝脏肿瘤细胞(HepG2)生长的抑制作用。结果壳聚糖纳米载体能显著增强端粒酶反义寡核苷酸对端粒酶活性和肿瘤细胞生长的抑制作用。结论壳聚糖纳米粒可作为反义技术治疗的有效载体。[Objective] To investigate the growth inhibition of antisense ohgonucleotides of telomerase on liver tumor cell enhanced by nanoparticles. [Methods] Chitosan nanoparticles were prepared and antisense oligonucleotides of telomerase were carried in them. Influence of antisense oligonucleotides of telomeras on telomerase was studied by TRAP-ELASA method. The inhibition of antisense oligonucleotides of telomerase on HEPG2 growth was studied by MTT method. [Results] Antisense oligonucleotides of telomerase carried by chitosan nanoparticles could enhance inhibition of liver tumor cell and telomerase. [Conclusion] Albumin nanoparticle can be used as an effective carrier in antisense technology.
