端粒酶反义寡核苷酸及其联合顺铂对胃癌细胞的影响

摘　　要：目的探讨端粒酶反义寡核苷酸及其联合顺铂对胃癌细胞的影响。方法利用脂质体将端粒酶反义寡核苷酸导入SGC7901胃癌细胞,TRAP-ELISA法检测端粒酶活性,噻唑蓝(MTT)法测定端粒酶反义寡核苷酸对胃癌细胞SGC7901的抑制;过氧化物酶的抗荧光素抗体法检测癌细胞凋亡指数。结果经端粒酶反义寡核苷酸处理的胃癌细胞端粒酶活性显著下降,端粒酶反义寡核苷酸明显抑制胃癌细胞的生长和诱导胃癌细胞的凋亡,端粒酶反义寡核苷酸联合顺铂,能增强对胃癌细胞生长的抑制和诱导胃癌细胞的凋亡。结论端粒酶反义寡核苷酸能有效抑制胃癌细胞生长,与顺铂协同作用,效果更为明显。[Objective] To study effect of telomerase antisense nucleotide and Cisplatin on gastric cancer cell, [Methods] Antisense nucleotide was transfected into cells by liposome, Telomerase activation were measu;ed through TRAP-ELISA. Influence of telomerase antisensenucleic acids on inhibition of gastric cancer cell was tested by MTT methods, Apoptotic index of tumor cell were determined by means of anti-dioxyfluoran antibody of peroxydase. [Results] After treated by antisense nucleotide, telomerase activation of gastric cancer cell decrease obviously. Antisense nucleotide of telomerase suppressed growth of gastric cancer cells and induced apoptosis of these cells significantly. When gastric cancer cells were treated with antisense nucleotide and Cisplatin, growth suppression and apoptosis were enhanced. [Conclusions] Antisense nucleotide of telomerase can effectually suppressed the growth of gastric cancer cells, and the suppression was reinforced when antisense nucleotide of telomerase and Cisplatin were used meantime.
