摘 要:目的探讨电离辐射影响人成纤维细胞基因表达的转录调控机制,并构建基因与相应转录因子的调控网络。方法先对前期研究已经获得的差异表达基因进行功能注释,选择结合功能类基因(数量最多)进入研究;再用MaxLAPS,TFME和SCOPE3种软件预测这些基因的转录因子和转录因子结合位点;利用Bibliosphere软件构建转录因子与基因的转录调控网络。结果获得25条GeneSymbols,基因本体功能注释集中于结合功能(共19条),2种程序预测得到14个相同的转录因子,得分最高的6个转录因子结合位点对应的转录因子与预测的基本一致,按照最高水平标准构建的转录调控网络,显示CASPASE-3位于网络的核心。结论电离辐射很可能经转录因子TP53介导CASPASE-3激活而致人成纤维细胞凋亡。Objective To explore the transcriptional regulatory mechanism in ionizing radiation-damaged human fibroblasts. Method Differentially expressed genes obtained from previous studies were functionally annotated and the genes under the binding function category were selected for further study. MaxLAPS, TFME and SCOPE programs were used to predict the genes encoding for the transcription factors (TFs) and transcription factor binding sites (TFBS). Bibliosphere was used to construct the regulatory network for these genes and the related TFs. Result A total of 25 differentially expressed genes were obtained, in which 19 genes were annotated with binding activity. 14 corresponding TFs were predicted by these programs. The top six TFBS and the corresponding TFs are in agreement with the prediction from these softwares. Caspase-3 appeared to be the hubs in this regulatory network. Conclusion Ionizing radiation probably induced apoptosis in human fibroblasts via TP53/caspase-3 pathway.
