摘 要:目的通过对低氧高二氧化碳环境中高原鼢鼠(Myospalax baileyi)在不同季节血清中乳酸(Lactate,LD)含量、乳酸脱氢酶(LDH)活力以及LDH同工酶谱的测定,探讨高原鼢鼠对低氧高二氧化碳的适应机制。方法用分光光度法测定LD的含量、LDH活力,用聚丙烯凝胶电泳法对不同季节血清LDH同工酶进行电泳。结果高原鼢鼠血清中LD含量、LDH活力及LDH同工酶谱发生明显的季节性变化的趋势。LD含量、LDH活力在随着春季、夏季、秋季不同季节的变化而显著减小(P<0.05),血清中LD含量在春季、夏季和秋季分别为(6.98±1.20)mmol/L,(6.65±1.80)mmol/L,(3.17±0.38)mmol/L;LDH活力在春季、夏季和秋季分别为(5 865.79±1 338.95)U/mg,(4 493.73±1 153.47)U/mg,(3 338.11±523.74)U/mg。高原鼢鼠血清中LDH同工酶谱在不同的季节有特殊的酶带,在春季LDH同工酶含有明显的5条区带,均着色深且带型比较宽,夏季除LDH3、LDH4、LDH5的区带着色变淡外,LDH1、LDH2酶带几乎消失。秋季变化更明显,除LDH5区带外,其他的酶带几乎消失。结论血清中LD含量、LDH活力以及血清中LDH同工酶谱随季节的变化而呈现明显变化的趋势。Objective To investigate the mechanism of plateau zokors( Myospalax baileyi )adaptation to the hypoxic-high carbon dioxide environment by assaying the lactate(LD)content, lactate dehydrogenase(LDH) activity, and spectrum of LDH isoenzymes in serum of plateau zokors. Methods The LD content and the LDH activity were determined by spectrophotometry. The LDH isoenzymes were analyzed by polyacrylamide gel electrophoresis. Results The LD content, LDH activity, and spectrum of LDH isoenzymes showed significant changes in different seasons. The LD content and LDH activity decreased sitmificantlv in soring, summer, and autumn ( P 〈 0.05). The LD contents in spring, summer, and autumn were 6.98±1.20 mmol/L, 6.65± 1.80 mmol/L, and 3. 17 ± 0. 38 mmol/L, respeetirely, whereas the LDH activities were 5 865.79± 1 338.94 U/mg,4 493.73 ±1 153.47 U/mg, and 3 338.11 ± 5 23.74 U/mg, respectively. The spectra of LDH showed specific bands in serum of plateau zokors in different seasons. The spectnma of LDH isoenzymes in spring contained five bands, i.e. LDH1, LDH1,LDH3, LDH4, and LDH5, which were deeply stained and broad. But the LDH1 and LDH2 in summer nearly disappered while the bands of LDH3, LDH4 and LDH5 were lightly stained and narrow.The bands of LDH isoenzymes in autumn nearly dissappered except for LDH5. Conclusions The LD content, the LDH activity, and the spectrum of LDH isoenzymes in serum of plateau zokors vary with seasons.
