恶性胸水中exosomes的分离和鉴定

摘　　要：为exosomes用于肿瘤的免疫治疗,用多步离心超滤和密度梯度离心方法从恶性胸水中分离exosomes,电镜、蛋白定量和Hsc73对exosomes进行分析鉴定。结果显示分离出exosomes的形态为圆形或椭圆形小囊泡,直径约3O^9O nm,在标记金颗粒的Hsc73作用后,有金颗粒附着。To use exosomes in immune therapy of tumour, exosomes were-separated by a sequence of differential steps centrifugation ultrafiltration and density gradient centrifugation from malignant pleural effusion, the results showed that exosomes was rounded or elliptic vesicles by electron microscope, quantitative study of protein and Hsc73, whose diameter were 30-9Ohm, and after reaction with Hsc73 labeled by nanoparticle gold, there were gold particle.
