摘 要:利用ISSR分子标记技术,对51个茶树品种进行遗传多样性分析。选用10个多态性高、分辨力强的ISSR引物分别对供试材料的基因组DNA进行扩增,共获得53条清晰可辨的带,其中多态性带49条,多态位点百分率为92.45%,表明供试品种资源在DNA水平上存在广泛的变异。51份资源所检出的位点平均有效等位基因数、平均基因多样度、平均Shannon信息指数,分别为1.621±0.333,0.351±0.162,0.514±0.219。遗传距离介于0.097~0.692之间,平均为0.337;UPGMA法将51份资源分成4个类群。亲缘关系树状图在分子水平上显示了供试茶树品种间的亲缘关系,为今后茶树育种亲本的选配提供了理论依据。Genetic diversity of 51 tea cuhivars [Camellia sinensis (L.) O.Kuntze] was revealed by ISSR-PCR (Inter-simple sequence repeat). A total number of 53 unambiguous bands were amplified from the 51 tea cultivars with 10 primers screened from 100 ISSR primers, 49 of which were polymorphic, and their polymorphism frequency was 92.45%. As analyzed by POPGENE 32, the average value of effective number of alleles, Nei's gene diversity and Shannon information index were 1.621±0.333, 0.351 ±0.162, 0.514±0.219, respectively. The Nei's genetic distances of the 51 tea cuhivars ranged from 0.097 to 0.692, with a mean of 0.337. The tested tea cultivars were divided into 4 groups by UPGMA (unweighted pair group method with arithmetic average) based on Nei's genetic distances. The genetic relationship of the 51 tea cultivars revealed by the ISSR dendrogram would be helpful for selecting the parents in tea breeding programs.
