摘  要:目的检测生存素(survivin)反义寡核苷酸对肝细胞癌细胞中生存素基因及蛋白表达的影响。方法将生存素反义寡核苷酸(ASODN)分为150、500和1000nmol/L3个不同浓度的亚组对SMMC-7721细胞进行转染,并设正义寡核苷酸(SODN)组、空脂质体组及空白组作为对照组。采用逆转录—聚合酶链反应(RT-PCR)方法检测各组中生存素mRNA的表达,采用流式细胞术(FCM)检测各组中生存素蛋白的表达。结果各ASODN亚组生存素mRNA及生存素蛋白的表达量均低于各对照组(P<0.05或P<0.01)。各ASODN亚组间比较,差异有统计学意义(P<0.05或P<0.01),并呈现剂量依赖性。SODN组、空脂质体组及空白组组间比较,差异均无统计学意义(P>0.05)。结论利用生存素反义寡核苷酸转染SMMC-7721细胞,可以下调生存素mRNA及其蛋白的表达。Objective To detect the expression of survivin mRNA and its protein, as the suvivin antisense oligodeoxynucleotide (ASODN) were transfected to the tumor cell of hepatocelluar carcinoma. Methods 150,500 and 1 1300 nmol/L survivin ASODN groups were transfected into SMMC -7721 ceils, And the control groups were defined as SODN group, Oligofectamine group and control group. Reverse transcription polymerase chain reaction (RT-PCR) was perfomaed to evaluate the survivin mRNA expression level in each group, Flow cytometry(FCM) was used to detect the level of survivin protein expression in each group, Results In ASODN groups, the expression level of survivin mRNA and protein decreased significantly compared with control groups ( P 〈 0.05 or P 〈 0.01 ), among each ASODN group there were significant differences ( P 〈 0.05 or P 〈 0.01 ). however no significant difference was found among each control group ( P 〉 0.05), Conclusion The expression level of survivin mRNA and protein in SMMC-7721 cells could be inhibited as the ASODN were transfected.
