摘  要:目的探讨提前光刺激对小鼠神经节细胞发育不同阶段感光视蛋白melanopsin表达的影响。方法对出生4dC57BL/6J小鼠右眼建立提前开睑接受光刺激模型,左眼作为自身对照,应用免疫荧光、RT-PCR和Western blotting方法比较小鼠5天龄、6天龄、7天龄、14天龄和90天龄时提前开睑组和对照组视网膜感光视蛋白melanopsin的分布及转录和翻译水平的表达变化。结果Melanopsin阳性细胞在5天龄时就已经在视网膜上明显表达,随着小鼠视网膜发育的逐渐成熟,提前开睑组和对照组melanopsin表达均逐步下降;组内比较,在6天龄、7天龄、14天龄时melanopsin提前开睑组表达量均少于各自对照组,差异具有显著性(P<0.05);90天龄时melanopsin表达最少,且提前开睑组和对照组差异无显著性(P>0.05)。结论小鼠14天龄之前是melanopsin阳性神经节细胞发育的关键阶段,在此阶段内,melanopain阳性神经节细胞数量逐渐减少,提前开睑光刺激使melanopsin阳性神经节细胞数量进一步减少。本研究结果有助于揭示melanopsin在视网膜神经节细胞发育过程中的表达变化及其可能的分子机制。Objective To investigate the effects of light stimulation prior to the expression of melanopsin during different develop- mental stages of retinal ganglion cells. Methods A model for light stimulation was established by opening the right eyelids of C57BL/ 6J mice at postnatal 4 days (P4), while the left eyes were used as a self-control. The distribution and expression of melanopsin were observed in the experimental and control groups at P5, P6, P7, P14 and P90 with immunohistochemistry, RT-PCR and Western blotting, Results The retina showed significant expression of melanopsin-expression cells at P5. As the development of the retina progressed, the expression of melanopsin was gradually down regu- lated. Expression in the experimental group was significantly lower than that in the control group before P14 (P〈0.05). However, there was no significant difference between the experimental and control groups on P90 (P〉0.05), Conclusion A key stage for the develop- ment of retinal ganglion cells occurs before P14. The total amount of melanopsin-expression cells gradually decreased, and introducing light stimulation by opening the eyelids can down regulate melanopsin expression. The results may be useful in providing insight on the expression of melanopsin during the development of retinal ganglion cells and possible molecular mechanisms.
