摘  要:目的:探讨胰岛素样生长因子Ⅰ受体(IGF-ⅠR)反义寡核苷酸(ASODN)对肺癌NCI-H460细胞中IGF-ⅠR蛋白表达的影响。方法:设计并合成3条IGF-ⅠR反义寡核苷酸序列,转染NCI-H460细胞,另设无关的IGF-ⅠR反义寡核苷酸序列和空白培养液作对照,细胞培养48h后,蛋白酶消化收集细胞,滴于预处理的载玻片上。采用免疫细胞化学SP法检测各组NCI-H460细胞中IGF-ⅠR的蛋白表达。结果:3条IGF-ⅠR ASODN转染的NCI-H460细胞中IGF-ⅠR蛋白表达差异无统计学意义(P>0.05),但均低于无关寡核苷酸转染组及空白培养液对照组(P<0.05)。结论:IGF-ⅠR ASODN可抑制肺癌NCI-H460细胞中IGF-ⅠR蛋白的表达。Aim : To investigate the effects of IGF- I R antisense oligonucleotide (ASODN) on protein expression of IGF- I R in human lung cancer NCI-H460 cell. Methods:Three IGF- I R ASODN sequences were designed and synthesized. The NCI-H460 cells were transfected by IGF- I R ASODN ,N-ODN. The expression of IGF- I R protein was detected by immunocytochemistry SP method. Results: The positive expression of IGF- I R protein was examined and could be in- hibited by ASODNs. There was no significant difference among 3 ASODN groups ( P 〉 0.05 ) , bat there was significant difference between ASODNs and N-ODN or non-transfected group( P 〈 0.05 ). Conclusion : The expression of IGF - I R protein in NCI-H460 cells could be inhibited by ASODNs.
