摘 要:选择了具有生物活性的壳聚糖为载体、戊二醛为交联剂来固定化α-淀粉酶,探索了固定化的最佳条件。结果表明:最佳固载量为每克壳聚糖50mgα-淀粉酶,交联剂戊二醛浓度为15%,处理温度均为室温,处理时间以5h为宜,固定化pH值以7较好。交联壳聚糖用于固定α-淀粉酶行之有效,且效率较高,其酶活回收率可达82.2%。On the condition of cross link by glutaraldehyde,the chitosan was chosen as the carrier of α-amylase due to its biocompatibility.The condition of the immobilization of α-amylase was studied.The optimum amount of enzyme immobilized on chitosan was 50mg/g chitosan.The optimum temperature and time of glutaraldehyde reacting with chitosan were ambient temparature and 5 hour respectively.The concentraion of the glutaraldehyde was 15%.The optimum pH value of the immobilized enzyme was 7.0 for α-amylase.This research certified that it was practicable and efficient that α-amylase was immobilized on chitosan with glutaraldehyde.The recovery of immobilized enzymes activity was 82.2%.
