摘 要:研究了蜂花粉中葡萄糖、果糖、蔗糖、麦芽糖的高效液相色谱(HPLC)分析方法。花粉样品用热水溶解,超声波提取,亚铁氰化钾溶液和乙酸锌溶液沉降蛋白,使用SUPELCO ENVITM固相萃取小柱去除色素等干扰成分后,以Waters Carbohydrate分析柱为色谱柱,乙腈-水(体积比79:21)为流动相进行分离,回收率90.4%~96.2%,精密度为2.4%~4.3%。利用本方法对8种花粉的80个样品进行了分析,发现花粉中果糖和葡萄糖的平均含量分别在10.9%~12.6%和6.8%~9.6%之间。该方法简便快捷,可作为标准方法对蜂花粉中葡萄糖、果糖、蔗糖和麦芽糖进行分析,是一种对花粉糖分组成进行质量控制的有效方法。A HPLC method with refractive index detection (RID) was developed for determination of sugars in bee pollen. Sugars were extracted from samples with water by ultrasonic oscillation and isolated by solid phase extraction with SUPELCO ENVITM cartridge after protein precipitation by using K4Fe(CN)6 and Zn(CH3COO)2. Sugars were separated on a Waters high performance carbohydrate column with acetonitrile-water(79:21, WV) as mobile phase. The relative standard deviations are 2.4%~4.3% and the recoveries are 90.4%~96.2%. The HPLC method was applied for the analysis of 80 bee pollen samples from 8 species. The average concentrations of fructose and glucose are 10.9%~ 12.6% and 6.8%~9.6%, respectively. The method is simple and suitable for routine quality control of bee pollen sugar composition.
