摘  要:[目的]观察茶多酚在甲醛损伤人内皮细胞(HUVEC-12)中的保护作用及其可能机制。[方法]体外培养HUVEC-12细胞,预孵茶多酚12h后加入甲醛作用4h,透射电镜观察细胞形态改变,分光光度法测定细胞培养液中丙二醛(MDA)含量、超氧化物歧化酶(SOD)活力及一氧化氮(NO)含量。[结果]预孵茶多酚组细胞形态损伤减轻,MDA、SOD、NO,2.5mg/L~20mg/L茶多酚各组与0.1mmol/L甲醛组比较,差异有统计学意义(P﹤0.05)。[结论]茶多酚可以拮抗甲醛对内皮细胞的损伤,其机制与茶多酚减少NO的产生,抑制细胞内脂质过氧化有关。[ Objective] To investigate the protective effects and mechanism of green tea polyphenols (GTP) against formaldehyde-induced injury of human umbilical vein endothelial ceils. [ Methods] Human umbilical vein endothelial cells (HUVEC-12) were cultured with DMEM in vitro and selected the ceils in the exponential phase of growth. The ceils were divided into control group, 0.1 mmol/L formaldehyde group and four GTP groups (2.5 mg/L, 5 mg/L, 10 mg/L, 20 mg/L). Various concentrations GTP were preineubated with cells for 12h before incubated with 0.1 mmal/L formaldehyde, then detected indexes mentioned below. The cell morphology was observed by transmission electron microscope (TEM). The concentrations of malondialdehyde (MDA), the activity of superoxide dismutase (SOD) and the level of nitric oxide (NO) were determined by spectrophotometric method. [Results] Formaldehyde could induce cell injury. The level of MDA and NO in FA group were significantly increased, comparing with control (P 〈 0.01 ). The activities of SOD in FA group were decreased significantly, comparing with control ( P 〈 0.01 ). The formaldehyde -induced alterations were completely prevented by pre-incubating the cells with GTP for 12h., comparing with 0.1 mmol/L FA group (P 〈 0.01 ). [Conclusion] GTP can alleviate formaldehyde-induced damages in dose-dependent manner. The protective mechanism may be GTP decrease the production of NO and alleviate the lipid peroxidation of HUVEC-12.
