摘  要:目的探讨缺氧诱导因子1α(HIF-1α)、基质金属蛋白酶-2(MMP-2)在膀胱移行细胞癌(BTCC)组织中的表达及其与临床生物学行为之间的关系。方法应用免疫组织化学检测BTCC组织中HIF-1α和MMP-2的蛋白表达,分析其表达与肿瘤部分临床病理特点的关系,并分析二者表达的相关性。结果正常膀胱组织中均未见HIF-1α和MMP-2蛋白表达,膀胱移行细胞癌组织中HIF-1α和MMP-2蛋白的阳性表达率分别为52.63%(30/57)和49.12%(29/57);HIF-1α阳性表达与肿瘤的临床分期及复发明显相关;MMP-2的阳性表达与肿瘤的病理分级、临床分期显著相关;HIF-1α的表达与MMP-2的表达密切相关。结论HIF-1α及MMP-2在BTCC的侵袭等生物学过程中具有重要的作用,HIF-1α和MMP-2也许可用于临床监控BTCC的进展。Objective To investigate the expressions of hypoxiainducibile factor lα( HIF - lα) and matrix metalloproteinase 2 ( MMP - 2) protein and their relationship with the biological behavior in bladder transitional cell carcinoma( BTCC). Methods HIF- lα and MMP- 2 protein expression were determined using inmmnohistoche- mistty. The relationship between HIF - lα, MMP - 2 expression and various clinicopathological parameters including pathological grade and clinical stage was analyzed and the correlation between HIF- l ctexpression and MMP - 2 expression was also analyzed. Results No marked expression of HIF - lα and MMP - 2 was observed in the normal bladder tissues. The positive rate of HIF - lα and MMP - 2 in BTCC was 52.63% (30 of 57) and 49.12% (29 of 57) respectively. HIF- 1α protein expression was significantly associated with patients, TNM stage and relapse( P 〈 0.05). Significant differences existed in the expression of MMP - 2 in carcinomas from different pothological grades and clinical stages (P 〈 0,05). The expression of HIF- lα protein was sig41ificantly correlated with MMP- 2 protein expression. Conclusion HIF- lα and MMP - 2 may play a critical role in the invasion of bladder carcinoma and may be used to estimate the progression of bladder carcinoma.
