摘 要:建立了环带斑褶菇菌丝体粗多糖的甲醇一步提取法。采用正交设计,优化了提取工艺,并采用传统的水提醇沉法验证了新工艺的应用效果。结果表明,甲醇一步提取法可以有效地应用于环带斑褶菇菌丝体粗多糖的制备。采用甲醇一步提取法提取环带斑褶菇菌丝体粗多糖的最佳工艺为:料液比1∶40,提取温度55℃,提取时间6h。甲醇一步提取法提取的粗多糖为白色半透明结晶,蛋白含量0.02%,得率23.46%;而水提醇沉法提取的粗多糖为浅褐色粉末,蛋白含量0.36%,得率22.04%。甲醇一步提取法有待于在其他生物粗多糖提取中得到更大范围的应用。A new technology of direct extraction by methanol was set up to prepare the crude polysaccharide from mycelia of Panaeolus cincutlus. Orthogonal design was used to optimize the extraction technology and the traditional method of hot water extraction was applied to verify the applicability of the new technology. The re- sults suggested that direct extraction by methanol was effectively applied in the preparation of crude polysaccharide from mycelia of Panaeolus cincutlus. The best technology was: ratio of dosage liqueur 40, temperature 55℃ and 6 hours of extraction time. The extracted crude polysaccharide using the new technology was white crystal with translucency and low (0.02 %) content of protein and high (23.46 %) extraction ratio. The crude polysaccharide extracted by hot water extraction and alcohol precipitation was beige powder with higher (0.36 %) content of protein and 22.04 % extraction ratio. The new technology of direct extraction using methanol will be further applied in crude polysaccharide extraction from other samples.
