摘  要:目的探讨实时超声检测野百合碱所致兔慢性肺动脉高压模型的建立及其意义。方法采用新西兰大白兔32只,实验对照组8只,其余用于建模,给予99%野百合碱溶液腹腔注射,同步实时超声检测,以右心导管压力测定值作为金标准,建立慢性肺动脉高压动物模型。结果24只兔中3只麻醉意外死亡,1只失血性休克死亡,成功13只,成功率65%。除了右心室舒张压外,模型兔各个心腔压力均较对照组显著增高(P<0.05)。模型兔干预后,肺动脉血流加速时间(AT)、射血时间(ET)显著缩短(P<0.05),而射血前期时间(PEP)及射血前期时间与加速时间比值(PEP/AT)则显著延长(P<0.05)。结论采用实时超声检测建立野百合碱所致兔慢性肺动脉高压模型简便易行,成功率较高,为慢性肺动脉高压的基础和临床研究提供了一个良好的平台。Objective To explore the establishment and significance of a rabbit model with chronic monocrotaline-induced pulmonary hypertension monitored by real-time echocardiography. Methods Chronic pulmonary hypertension in a rabbit model was developed using 99% monocrotaline solution injection into abdominal cavity, and synchronously monitored on real-time echocardiography. Then the pulmonary artery systolic pressure was detected by putting pressure-guided micro catheter put into the pulmonary artery (〉 30 mm Hg was the criterion of pulmonary hypertension ) .Results Of 24 healthy New Zealand rabbits, 13 rabbit models with chronic monocrotaline-induced pulmonary hypertension were successfully developed except 3 rabbits died of anesthetic accident and 1 rabbit died of hemorrhagic shock. The successful rate was 65%. In contrast to the control group, the pressure increase cardiac chambers were observed. Decrease in the AT and ET except the right ventricular diastolic pressure increase in the PEP and PEP/AT of the PA flow and decrease in the E/A ratio of the right ventricular inflow spectra, were all observed as pulmonary hypertension was developed. Conclusions This rabbit model with chronic monocrotaline-induced pulmonary hypertension is easy to be operated and the successful rate is high, which may provide a good way in the preclinical and clinical research.
