摘 要:通过丙酮分级沉淀、DEAE-纤维紊离子柱层析和Sephadex G100柱层析得到两种香菇漆酶同工酶,分别命名为lac1和lac2,并对其理化性质进行了研究。考察了香菇漆酶同工酶最适反应温度和pH值、热稳定性和pH值稳定性、反应动力学常数以及金属离子对酶活的影响。试验结果表明:Lac1、lac2最适作用温度都为30℃,在不高于40℃的条件下保存30min,lac1残余酶活在60%以上,lac2在不高于60℃有较高热稳定性,残余酶活在80%以上。最适pH值,lac1为3.6,lac2为4.0。pH值3.0-4.8,lac1有较强稳定性,在pH值4.0-4.6,lac2有较强的稳定性。以邻联甲苯胺为底物lac1和lac2的Km分别为0.964mmol/L和1.38mmol/L。金属离子Cr对lac1有激活作用,Ag^+、Mn^2+对lac1有抑制作用。金属离子Ba^2+和Cu^2+对lac2有激活作用,Fe^3+、Cl^-、Ca^2+、Zn^2+、Ag^+、Mn^2+对lac2有抑制作用。The laccase of Lentinus edodes was purified by classification deposition of acetone, DEAE - Cellulose ion exchange and Sephadex G100 column chromatography. Two sorts of laccases named lael and lac2 were acquired. We also studied the partial enzymological properties. The optimum temperature oflacl and lac2 were 30℃, and over 60% activity of the lacl remained within 30 min when the temperature was not higher than 40℃. The lae2 showed higher stability and over 80% activity of the lac2 remained and when the temperature was not higher than 60℃. The optimum pH of lacl and lac2 were 3.6 and 4. 0 respectively. The lacl was kept strongly active when pH ranged from 3.0 to 4.8. lac2 was 4.0-4.6. The Km of lacland lac2 were 0.964 mmol/L and 1.38 mmol/L respectively with o-Tolidine as substrate. The lacl was activized by G-and its activity was inhibhed by Ag^+ ,Mn^2+. The lac2 was activized by Ba^2+ and Cu^2+ and its ac- tivity was inhibited by Fe^a+ ,CI^-,Ca^2+ ,Zn^2+ ,Ag^+ ,Mn^2+.
