摘 要:【目的】探讨多聚(ADP-核糖)聚合酶(PARP)抑制对小鼠结肠癌CT26细胞体外运动侵袭能力的影响及机制。【方法】细胞运动及侵袭试验观察PARP抑制对CT26细胞运动侵袭能力的影响;Western Blot和明胶酶谱法分别检测PARP抑制对CT26细胞基质金属蛋白酶MMP-2、MMP-9表达和活性的影响。【结果】5-AIQ处理组CT26细胞运动迁移率(70%±7%)和侵袭率(63%±10%)较5-AIQ未处理组(100%±7%、100%±11%)均减弱(P<0.001)。5-AIQ处理组CT26细胞MMP-2、MMP-9表达(64%±12%、72%±12%)较5-AIQ未处理组(分别为100%±11%,100%±21%)明显减弱(P=0.0003、0.0163)。且CT26细胞培养上清液中MMP-2、MMP-9活性,在5-AIQ处理组[积分吸光度(IA)分别为0.34±0.07,0.40±0.05]与未处理组(0.48±0.10、0.61±0.08)之间同样存在明显差别(P=0.0248、0.0013)。【结论】实验结果提示,PARP抑制剂5-AIQ可降低CT26细胞的运动及侵袭能力,其可能与5-AIQ抑制PARP进而降低CT26细胞肿瘤侵袭转移相关因子MMP-2和MMP-9的表达和活性有关。[Objective] To approach the effects of poly(ADP-ribose) polymerase (PARP) inhibition on the migration and invasion of mouse colon carcinoma cell line (CT26) in vitro,and its mechanism. [Methods] The effects of PARP inhibition on migratory and invasive potencies of CT26 were observed by cell migration assay and invasion assay. The effects of PARP inhibition on expressions and activities of MMP-2 and MMP-9 were investigated by Western Blot and zymography, respectively. [ Results] The migratory rate (70% ± 7%) and invasive rate (63% ± 10%) of 5-AIQ-treated CT26 cells were both reduced (P 〈 0.00 1) compared with 5-AIQ-untreated cells (100% ± 7% and 100% ± 11%, respectively). The expressions of MMP-2 and MMP-9 of CT26 cells (64%±12% and 72% ± 12%, respectively) were significantly attenuated (P = 0.000 3 and P = 0.016 3) by 5-AIQ compared with 5- AIQ-untreated cells (100% ± 11% and 100% ± 21%, respectively). Moreover, the activities of MMP-2 and MMP- 9 both had significantly differences(P = 0.0248 and P = 0.0013) between supernatant of 5-AIQ-treated CT26 cells (IA : 0.34 ± 0.07 and 0.40± 0.05, respectively) and 5-AIQ-untreated cells (IA : 0.48 ± 0.10 and 0.61 ± 0.08, respectively). [Conclusion] The data suggested that PARP inhibitor 5-AIQ could inhibite migratory and invasive potencies of CT26 cells,probably because 5-AIQ decreased invasion-relative factors, MMP-2 and MMP-9 by inhibiting PARP of CT26 cells.
