摘 要:目的探讨高强度超声治疗对荷U14宫颈癌小鼠脾细胞Th1、Th2亚群细胞因子mRNA表达水平的影响。方法实验分为4组:HIU治疗组(A)、手术治疗组(B)、荷瘤对照组(C)及正常对照组(D)。于种植肿瘤后7 d,分别接受相应治疗;17 d,制备并培养小鼠脾淋巴细胞,应用逆转录多聚酶链式反应(RT-PCR)技术检测A、B、C、D各组小鼠脾淋巴细胞细胞因子mRNA的表达水平。结果A组小鼠脾淋巴细胞IL-2 mRNA相对含量(0.752±0.101)较B(0.430±0.064)、C(0.188±0.049)、D(0.392±0.053)各组明显增高(P<0.05)。A组小鼠脾淋巴细胞IFN-γmRNA相对含量(0.507±0.076)较B(0.233±0.045)、C(0.087±0.010)、D(0.218±0.049)各组明显增高(P<0.05);A组小鼠脾淋巴细胞IL-4 mRNA相对含量(0.213±0.049)较C(0.745±0.067)、D(0.310±0.064)各组明显降低(P<0.05);A组小鼠脾淋巴细胞IL-10 mRNA相对含量(0.093±0.032)较B(0.231±0.053)、C(0.657±0.111)、D(0.308±0.086)各组明显降低(P<0.05)。结论HIU固化的U14肿瘤细胞可能激活机体免疫功能,促使Th2占优势向Th1占优势逆转,从而提高小鼠抗肿瘤免疫功能,抑制肿瘤生长。Objective To investigate the effect of high intensity ultrasound (HIU) on the cytokine mRNA of T helper type 1 (Th1) and T helper type 2 (Th2) splenocyte subgroups from mou.se bearing U14 cervical cancer cells. Methods Twenty four mice bearing U14 cervical cancer cell were divided into four groups: HIU group (A), surgery resection group (B), no treatment group (C) and normal group (D) .On the seventh day after tumor transplantation, the mice in the four groups received different therapy. On the seventeenth day after tumor transplantation, reverse transcription polymerase chain reaction (RT-PCR) technique was used to study the levels of cytokine mRNA expression in splenocytes from tumor-bearing mice in group A/B/C and D. Results IL-2mRNA expression levels of mice splenocytes in group A (0.752±0.101) were higher than those in group B (0.430±0.064), groupC (0.188±0.049) and group D (0.392±0.053) (P〈0.05); IFN 7mRNAexpression levels of mice splenocytes in group A (0.507±0.076) were higher than those in group B (0.233±0.045), group C (0.087± 0.010) and group D (0. 218±0. 049) (P〈0.05); IL-4 mRNA expression levels of mice splenocytes in group A (0.213±0.049) were lower than those in group C (0.745±0.067) and group D (0.310±0.064) (P〈0.05); IL- 10 mRNA expression levels of mice splenocytes in group A (0. 093±0. 032) were lower than those in group B (0.231± 0. 053), group C (0. 657±0. 111) and group D (0. 308±0. 086), P〈0.05. Conclusions HIU solidified cancer cells may activate the transformation from Th2 to Th1, regulate the balance of Th2 to Th1 type cytokine, play an important role in increasing immune function of tumor bearing body.
