摘  要:目的探讨葛根中抗氧化活性部位的提取与分离,并寻找最佳提取方案。方法采用Fenton-紫外比色法比较水洗脱部位、70%乙醇洗脱部位以及总提取部位抗氧化活性的强弱。选用L9(34)正交试验,优选提取条件。结果预实验中各因素对提取分离结果均有影响,最好的提取分离方法是:粉碎后热回流,大孔树脂柱层析,70%乙醇洗脱。抗氧化活性依次是:醇洗脱部位>总提取部位>水洗脱部位。正交试验最佳工艺是:采用8倍于原料的70%乙醇,回流提取3次,每次1.5 h。结论该实验证明黄酮成分有明显的抗氧化活性,所选的最佳工艺稳定、简便、可行。Objective To investigate the extraction and isolation of antioxidative principle from Radix Puerariae lobatae and probe into the best extraction plan. Methods By Fenton ultraviolet spectrophotometry, the antioxidative activity were compared among the rinsed part by distilled water, the rinsed part by 70 % ethanol and total extraction part. The L9(3^4) orthogonal experimental design was used to optimize the extraction condition. Results The factors in the preliminary test had obvious influences on the extraction and separation. The best extraction and separation method was as follows: hot reflux after crushing, purifying by macroporous adsorption resin and eluting with 70 % ethanol. The activity of antioxidation was the rinsed part by 70 % ethanol 〉 total extraction part 〉 the rinsed part by distilled water. The optimal technological condition was as follows: extraction with 8 times of 70 % ethanol at 100 ℃ for 3 times (1.5 hours each time). Conclusion This study confirms the flavones have evident antioxidative activity and the optimal technological condition is stable, simple and feasible.
