摘  要:目的鉴定人类T淋巴细胞受体β链基因(Tcell receptor beta-chain,TCRβ)上两个毗邻基因,即胰蛋白酶原基因(cationictrypsinogen,PRSS1)与TCR Vβ7.2片段之间的间隔序列在胰腺炎和健康人群中的分布情况。方法对来自3位胰腺炎患者及2位健康体检者的DNA样本利用特异引物进行聚合酶链反应(PCR)扩增,产物经电泳、纯化后直接测序,并分析该序列的碱基在胰腺炎组和正常人群中的组成情况。结果利用自行设计的引物在2份胰腺炎患者外周血DNA样本中扩增出片段长度约为2.6 kb的条带,DNA测序结果显示两个毗邻基因之间的片段在健康人群和胰腺炎人群中均不是稳定组成。结论TCRβ序列中胰蛋白酶原基因(PRSS1)与TCR Vβ7.2基因之间间隔序列存在不稳定性。Objective To analyze the pancreatitis patients and healthy people distribution condition of the intervening seq (TCRβ) uence between PRSS1 genes and TCRβ7. 2 which adjoin in the homo sapiens T cell receptor Beta-chain Methods Polymerase chain reation by specific primer was performed to amplify the genomic DNA of 3 patients with pancreatitis and 2 heahhy adults. The PCR products were purified after electrophoresish with the purified product. We performed sequencing, then determined the characteristics of intervening sequence in the pancreatitis patients and healthy adults. Results The genomic DNA extract from peripheral blood of 2 panereatitis patients was amplified by the primer designed. It was amplified into one long DNA fragments about 2. 6 kh. DNA sequencing revealed that the compose of the segments between these two adjoining genes were variable among the healthy people and the pancrea- titis patients. There were many types of basic group or segments insert or lost in these regions. Conclusion The inter- vening sequences between PRSS1 gene and TCRβ7.2 gene in TCRβcluster regions are instability. The study of these intervening sequences will make a progress to the anthropology
