摘  要:本研究观察了杂色曲霉素(sterigmatocystin,ST)对小鼠Purkinje细胞TNF-α和TGF-β2 mRNA表达的影响。给予BALB/c小鼠一次灌胃ST(3000μg/kg),分别于灌胃后0.5、1、2、4、8和16h处死动物,用原位杂交方法观察了Purkinje细胞TNF-α和TGF-β2 mRNA的表达。TNF-α和TGF-β2mRNA在对照组和处理组各时间点的Purkinje细胞均有阳性表达。但各处理组与对照组比较,TNF-α和TGF-β2 mRNA表达显著增加(P<0.01)。上述结果说明单次ST灌胃后TNF-α和TGF-β2 mRNA在Purkinje细胞表达明显增加,TNF-α和TGF-β2在ST对脑组织的损伤过程中起着重要的作用。To investigate the effects of sterigmatocystin (ST) on mRNA expressions of TNF-α and TGF-β2 in Purkinje cells of mice, BALB/c mice were treated intragastrically by garage with ST at 3000 μg/kg. The expressions of TNF-α and TGF-β2 mRNA in Purkinje cells of mice were detected by in situ hybridization 0.5, 1,2, 4, 8 and 16 h after oral administration, respectively. The results showed that TNF-α and TGF-β2 mRNA expressions could be found in some Purkinje cells at every time point in both control and treated groups. The positive rate and intensity of TNF-α and TGF-β2 mRNA expressions were all significantly higher than those in control group ( P 〈 0. 01 ). The present results suggest that TNF-α and TGF-β2 mRNA expression were all significantly higher in Purkinje cells after oral ST. TNF-α and TGF-β2 might play a critical role in the injury of brain by ST.
