摘  要:目的:研究去甲斑蝥素对人膀胱肿瘤T24细胞生长抑制作用,并探讨其作用机理。方法:将含不同浓度去甲斑蝥素的RMPI-1640与T24细胞一起培养,分别在12h、24h、36h、48h、60h收集细胞进行MTT实验,计算细胞生长抑制率并筛选最佳药物浓度;用最佳浓度的药物与细胞共同培养,免疫印迹法检测作用不同时间后bcl-2和Bax表达变化,分光光度法检测Caspase-3、-8、-9活性变化。结果:去甲斑蝥素在体外对T24细胞增殖有明显抑制作用,其最佳浓度为60μmol/L;去甲斑蝥素(60μmol/L)与T24共同培养,随着作用时间延长Bax表达逐渐增强,bcl-2表达逐渐减弱;并且Caspase-3、-8、-9的活性均增加。结论:去甲斑蝥素在体外对T24细胞有良好的抑制作用,其作用机理可能与其激活Caspase-3、-8、-9活性,增强Bax和减弱bcl-2表达而诱导凋亡有关。Objective To study the effect of Noreantharidin(NCTD) on the inhibition of the human bladder carcinoma cell(T24) and the mechanism. Methods MTT assay was used to calculate inhibition ratio when cells were treated with different concentration of NCTD for 12h, 24h, 36h, 48h and 60h. After treating cells with optimization density NCTD, the changes of expression of bcl - 2 and Bax were detected using western blotting and the active variation of caspase - 3, - 8 and - 9 was detected using spectrophotometeric,method. Results Proliferation of T24 cells was inhibited by NCTD and its optimization density was 60 μmol/L. The expression of Bax of T24 cells was enhanced and the expression of bcl - 2 was tapered gradually by NCTD. Furthermore, the activities of Caspase - 3, - 8 and - 9 were increased. Conclusion NCTD can inhibit the growth of T24 and the mechanism may be related to the increasing expression of Bax and the weakening expression of bcl - 2, beside the activation of Caspase - 3, - 8 and - 9.
