摘  要:目的:建立脊髓脊膜膨出(myelomeningocele,MMC)胎鼠模型并研究畸形机体抗氧化能力的改变。方法:取妊娠小鼠40只,随机分为3个实验组和1个对照组,分别于不同的时间点连续灌服全反式维甲酸(all-trans retinoic acid,ATRA)和同剂量大豆油,于孕18日剖宫取胎:①观察胎鼠外观畸形情况、椎板闭合情况、脊髓病理及神经细胞超微结构改变,②测定畸形胎鼠肝组织内超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GSH-Px)、总抗氧化能力(total antioxi-dant activity,T-AOA)及丙二醛(malonyl dialdehyde,MDA)含量。结果:实验组共获得MMC胎鼠93只,总畸形率达61.2%,各组MMC畸形率分别为T1-37.2%、T2-68.3%、T3-73.9%;各实验组GSH-Px、SOD、T-AOA均低于对照组(P<0.05),MDA含量均高于对照组(P<0.05)。结论:ATRA致畸是制作MMC胎鼠模型的简单有效的方法,MMC胎鼠抗氧化机能存在不同程度的降低。Objective:To establish myelomeningocele (MMC) mouse model and study body antioxidative activity in deformed fetus. Methods:40 pregnant mice were chosen and randomly divided into 3 test groups and 1 control group. To develop MMC mouse model, the consecutive giving o-rally of all-trans retinoic acid (ATRA) (12.5mg/kg.body weight) to pregnant mouse of test groups on 8 days of gestation at different time were performed. The animals of control group were only given equally the doses of soybean. All dams were sacrificed on 18 days of gestation, the uterus removed for the following: ①Vertebrae malformations, histological studies of spina code nerve in MMC mouse by light microscopy observations and inspection of unusual spinal neuronal ultrastructure. ②Detection of MDA、SOD、GSH-PX、T-AOA in Liver tissue of live fetus from each group. Results:There were total 93 MMC fetus harvested from 3 test groups. The total MMC incidence was 61.2% and the incidence in T1、T2 and T3 was37.2%、68.3% and 73.9% individly. The GSH-Px、SOD、T-AOA of 3 test groups were all lower than that of control group (P< 0.05) and the MDA of 3 test groups were higher than that of control group ( P < 0.05) . Conclusion: It is a good selection to establish animal model of MMC in mouse by consecutive giving orally of ATRA. There are low body antioxidative activity in fetus with MMC defect.
