摘  要:研究以羧甲基纤维素作为载体,通过接枝后与胰凝乳蛋白酶进行偶联,通过正交实验测得酶固定化特性的最佳方案为:接枝温度50℃,接枝时间10min,酶偶联温度20℃,酶偶联时间4h。所得固定化酶的热稳定性提高5℃,最适pH提高到8~9,具有连续操作性,在2℃贮存放置20d后酶活仍在75%以上。A method was proposed to immobilize chymotrypsin on carboxymethyl cellulose. The characteristics of the immobilized enzyme was discussed and assured the best condition of immobilized enzyme by orthogonal experiment. The results showed that the best condition was 50℃ and 10min of arms-on-carboxymethyl cellulose, 20℃ and 4h of immobilized enzyme. Its heat-stability was higher 5℃ than the native, and the optimum pH reached to prig-9. Also its operational stability and storage stability were better. Its activity had over 75% staying for 20d at 2℃.
