摘 要:[目的]研究超声波辅助提取羊肚菌多糖的条件。[方法]以水作溶剂,超声波辅助提取发酵培养的羊肚菌菌丝体中的多糖,研究提取时间、提取温度、提取次数、超声波作用功率、料液比对多糖提取率的影响。[结果]超声波作用功率为400 W时,多糖提取率最高。随着提取时间的延长和料液比的增大,多糖提取率先上升后下降。超声波辅助提取羊肚菌多糖的最佳条件为:料液比1∶10(g/ml),超声波作用功率400 W,提取时间0.75 h,提取温度55℃,提取次数2次。在该条件下,羊肚菌多糖提取率达0.786 2%,且所得多糖纯度较高,多糖含量为72.81%,多糖中蛋白质含量为0.92%。[结论]超声波辅助提取羊肚菌多糖具有时间短、节省能源、提取率高等优点。[Objective] The aim was to study the condition for ultrasonic wave assisted extraction of polysaccharide from Morchella esculenta.[Method] With the water as solvent,the polysaccharide from M.esculenta mycelium through fermentation culture was obtained by ultrasonic wave assisted extraction.The influences of extraction time,extraction temperature,extraction times,ultrasonic wave power and solid-liquid ratio on polysaccharide extraction rate were studied.[Result] The polysaccharide extraction rate was highest when the ultrasonic wave power was 400 W.The polysaccharide extraction rate was first increased and then decreased with the prolonging of the extraction time and increase of the solid-liquid ratio.The optimum condition for ultrasonic wave assisted extraction of polysaccharide from M.esculenta was solid-liquid ratio of 1∶10(g/ml),ultrasonic wave power of 400 W,extraction time of 0.75 h,extraction temperature of 55℃,extraction times of 2.Under the condition,the extraction rate of polysaccharide from M.esculenta was 0.786 2% and the purity of obtained polysaccharide was higher,the polysaccharide content was 72.81% and the protein content in polysaccharide was 0.92%.[Conclusion] Ultrasonic wave assisted extraction of polysaccharide from M.esculenta had some advantages such as short time,saving energy sources and higher extraction rate.
