摘 要:细胞毒细胞杀伤肿瘤细胞主要通过细胞凋亡机制。用传统的MTT法探讨了肿瘤浸润淋巴细胞TIL细胞杀伤活性最佳条件,同时采用凋亡荧光染色法(AFS)检测TIL的生物学效应。结果发现AFS法比MTT法更能准确地反映TIL对靶细胞的杀伤,杀伤率与效靶比呈良好的线性关系(r=0.89);且该法简便、快速、敏感、重复性好,能同时观察活细胞、凋亡及坏死三种细胞的形态特征。The apoptotic cells may be induced by the cytotoxicity cells. The authors detected the cytotoxicity of TIL by using apoptosis fluorescence staining. A comparision between the results of AFS assay with the results of MTT assay showed signigicant difference. There was a high degree of over a wide range of E/T ratios and cytotoxicity percentage (the r vaule is 0. 89) in AFS assay, while that of MTT assay showed no linear relationship. We found that the AFS as- say was a simple, rapid, accurate, sensitive and reproducible method to detect cytotoxicity and avoided different affect- ing factors of the MTT method. In addition, this new approach can differetiate concurrently whether these cells belong to apoptosis, necrosis or live cells.
