摘 要:目的分析RhoC及其调节蛋白GDP解离抑制因子α(Guanine dissociation inhibitor,GDIα)在肺癌细胞中的表达及其与肺癌细胞转移能力间的关系。方法应用Westernblot、RT-PCR分别检测正常支气管上皮细胞、不同的肺癌细胞系中的RhoC、Rho-GDIa蛋白及mRNA的表达。结果RhoC、Rho-GDIα在人支气管上皮细胞、肺腺癌细胞系、肺巨细胞癌细胞系均有表达,免疫荧光显示均表达于细胞浆。RhoC、Rho-GDIα在肺癌中的表达高于人支气管上皮细胞。在高转移能力的肺巨细胞癌亚系BE1RhoC、Rho-GDIα的表达均高于低转移能力的肺巨细胞癌亚系LH7。结论RhoC、RhoGDIα在肺癌细胞系中过表达并与转移能力相关。Objective To analyze the expression of Rhoc and Rho-GDIα in lung cancer cell lines and their correlation with metastatic abilities. Methods We used Western blot and reverse transcriptase polymerase chain reaction (RT-PCR) to detect the expression of RhoC and Rho-GDIα in normal bronchial epithelial cells and lung cancer cell lines. Results Both protein and mRNA of RhoC and Rho-GDIα were expressed in human normal bronchial epithelial cell, lung adenocarcinoma cell line and lung giant cell carcinoma cell line. Immunofluorescent staining showed that the expression was in the cytoplasma. The expression of RhoC and Rho-GDIα were higher in the cell lines than that in normal bronchial epithelial cells, and higher in the BE1 subclone of lung giant cell carcinoma cell line with high metastatic capacity than that in LH7 with low metastatic capacity. Conclusion RhoC and Rho-GDIα are overexpressed in lung cancer cell lines and may be correlated with metastasis.
