摘  要:以内蒙古和青海等地区牛乳中分离的11种野生乳酸菌为材料,分别应用核酸电泳技术、药物敏感试验和SDS-变温质粒DNA消除等方法,进行了内源性质粒DNA的普查、消除及其稳定性试验。结果表明:11种野生乳酸菌均被检测出不同大小的质粒DNA的可疑条带,可分为大于23 kb和小于23 kb两个型;经SDS-变温方法消除质粒DNA后,除3种乳酸菌不宜生长而被淘汰以外,在剩余8种乳酸菌中,小于23 kb型的条带全部被消除,个别大于23 kb型的可疑条带也能被消除,与药物敏感补充试验所得到的结果基本相符;已被消除质粒DNA的8种乳酸菌经过转接培养20代,显示出良好的遗传稳定性。本研究为筛选和建立新型基因工程受体菌奠定了基础。In this study, 11 species of lactic acid bacteria were isolated from milk samples, which were collected from Inner Mongolia and Qinghai. The general investigation, elimination and stability of the endogenous plasrnid DNA of these bacteria were carried out by nucleic acid electrophoresis, medicine sensitive test and SDS-variable temperature elimination, The results showed that all of the 11 species had the suspi- cious bands of plasmid DNA in different sizes. And these bands could be divided into two types, the one was larger than 23 kb and the other was smaller than 23 kb. 3 species were fallen into disuse because they could not grow after their plasmid DNA was eliminated by SDS-variable temperature elimination. In the residual 8 species of lactic acids bacteria, the smaller bands were completely eliminated by the method of SDS- variable temperature elimination and a few of the larger bands could be eliminated too. The above results corresponded with the medicine sensitive subsidiary test. The 8 species of lactic acid bacteria that had been eliminated the plasmid DNA presented the good hereditary stability after being cultured 20 generations. This study has laid the foundation for the screening and establishing new genetic engineering host.
