摘 要:分别研究了3种诱导剂和Mn2+浓度对重组人锰超氧化物歧化酶(rhMn-SOD)发酵表达的影响,并进一步优化了乳糖诱导rhMn-SOD表达的条件。结果表明:异丙基--βD-硫代半乳糖苷(IPTG)和乳糖诱导能够获得类似的表达效果,诱导的同时加入2 mmol/L的Mn2+能够明显提高rhMn-SOD的表达。进一步的实验结果表明:乳糖诱导的最佳浓度为7 mmol/L,最佳时机为接种后6 h,A600为2.1,诱导表达6 h rhMn-SOD活性和比活达到最高。The effects of three kinds of inducers and different concentrations of Mn^2+ on the expression of recombinant human manganese superoxide dismutase (rhMn-SOD) were studied. The inducement conditions of lactose on the expression of SOD were further optimized. Results showed that IPTG and lactose had similar effects on the expression of rhMn SOD. Adding 2 mmol/L of Mn^2+ at the time of inducement improved the expression of rhMn-SOD. Further studies indicated that the suitable inducement conditions were addition of 7 mmol/L lactose at 6 h after inoculum, and the cell optical density A600 could reach 2.1.
