摘 要:目的探讨髓过氧化物酶(MPO)G-463A基因多态性与大肠癌易感性的研究。方法取103例大肠癌组织和143例癌旁正常大肠组织(对照组)的石蜡切片样本,应用聚合酶联反应(PCR)及限制性片断长度多态性(RFLP)进行检测。结果MPO基因型(GG、GA、AA)在大肠癌组的分布为47.57%、31.07%、21.35%,对照组为27.97%、37.06%、34.97%,两组间比较差异有统计学意义(P<0.05);含MPO G-463A等位片段的基因型在大肠癌组中的分布为40.0%,对照组为52.55%,两组间比较差异有统计学意义(P<0.05)。结论MPO G-463A基因多态性可能是大肠癌发病的一个基因型危险因素,MPO基因463位点的A等位基因在大肠癌组中的表达较高,可能具有对肿瘤进行早期诊断和判断预后的潜在价值。Objective To investigate whether the gene polymorphism of myeloperoxidase(MPO) G-463A confers the susceptibility to large intestine cancer. Methods The tissue specimens of 103 patients with large intestine cancer and 143 cases of normal tissue besides the cancer sites were analyzed by the polymerase chain reaction(PCR) and restriction fragment length polymorphism(RFLP). Results The rates of MPO genotypes GG, GA, AA were 47.57% , 31.07%, 21.35% respectively in adenocarcinomas and 27.97% , 37.06% , 34.97% in the normal mucosa respectively. The differences were significant in the two groups(P 〈 0.05). The frequency of the A allele in adenocarcinomas was higher than that in control group(P 〈 0.05 ). Conclusion The MPO G-463A gene polymorphism may be one of the genetic risk factors for the development of large intestine cancer. The frequency of the A allele in adenocarcinomas is higher, which may play an important role in the development and prognosis of large intestine cancer.
