摘  要:目的:观察重组逆转录病毒的产生及感染K562细胞的超微结构。方法:将含neo基因的逆转录病毒载体导入人红白血病细胞系K562后,用电子显微镜观察。结果:细胞的胞体显著增大,核/浆比值降低,高尔基氏体显著增生、扩大,胞质内出现大量包涵体样结构,内质网复合体专一性地出现于转导细胞的胞质内,同时观察到细胞膜表面的芽生病毒颗粒,感染的K562细胞发生进行性死亡。结论:重组逆转录病毒载体在包装过程中产生了有复制能力的逆转录病毒,它们可在转导的K562细胞中复制、合成、装配,导致细胞代谢失衡及细胞死亡。Objective:To observe replication of recombinant retrovirus and ultrastructure of infected K562 cells. Methods:Retroviral vector containing neo gene was transduced into human erythroleukemia cell line K562. Transduced K562 cells were examined morphologically by electron microscopy. Results: There was an increase of cytoplasmic volume,particularly concerning Golgi apparatus. Numerous inclusion like structures aggregated in Golgi region. Annulate lamellae appeared exclusively in the infected K562 cells. Budding retroviral particles were found. Infected K562 cells were destructed progressively. Conclusion: The recombinent viruses can replicate and assembly in the cytoplasm of K652 cell after gene transfer by retrovirus vector. The replication competent retrovirus,a highly virulent strain, was produced through recombination during packaging.
