摘 要:目的:探索简单实用重复性好的犬肺静脉肌袖和心房肌细胞的分离方法。方法:采用两步酶解法,分离得到犬肺静脉肌袖细胞和心房肌细胞,用形态学和台盼蓝染色鉴别分离的心肌细胞,使用倒置显微镜观察和摄像记录。结果:分离心肌细胞的存活率为30%~60%,形态呈杆状、折光性好、横纹清晰,膜状态良好。结论:使用两步酶解法能获得存活率高,数量较多的心肌细胞,是一种简单实用的分离心肌细胞的方法。Objective: To explore an easier, more practical and reliable technique for the dial sleeves and atrial myocytes from canine. Methods: The two steps enzymatic diss isolation of myocar-ociation method was used to yield living cell from atrial and pulmonary vein sleeve tissue. The cardiomyocytes were morphologically observed with microscope and evaluated through trypan blue dying. Results.. Viability of freshly isola- ted adult canine myocardial sleeves and atrial myocytes, which were rod-shaped with clear cross-striations, was 30%-60%. Conclusion: The procedure of isolation used the two steps enzymatic dissociation was more convenient and reliable and can produce large numbers of single cardiamyocytes with high survival rate.
