摘  要:以抗旱马铃薯品系H154的试管苗为材料,采用液氮研磨,用Trizol试剂提取马铃薯总RNA,并进行分析,电泳检测得到多条清晰的谱带,且28 S和18 S条带的亮度比例接近2∶1;OD260/OD280平均值为1.91,浓度平均值为4.65μg/μL。结果表明,总RNA完整性强、纯度高、含量高,为进行马铃薯抗旱相关基因的克隆和分析奠定了一定的研究基础。The total RNA extracted from seedlings in vitro of potato line H154 with good dry resistance by Trizol method was analyzed by gel electrophoresis. The results showed that some clear spectral bands were detected, the brightness ratio between 28S and 18S band was near 2 : 1, the average value of OD260/OD280 was 1.91 and its concentration average value was 4.65 g/L The extracted Total RNA with good completeness, high purity and high content can provide the basis for studying cloning and analysis of related genes with dry resistance in potato to a certain degree.
