摘 要:本实验研究了应用琼脂扩散法测定饲料中所含木聚糖酶的活性。结果表明,该方法标准曲线制作线性良好;应用淀粉稀释木聚糖酶进行验证性实验,检测酶活测定值和实际理论值之间差异不显著(P>0.05),说明该方法准确率很高;应用饲料稀释木聚糖酶进行验证性实验,检测酶活值低于理论值,但这并不影响该方法对饲料中酶活的判定。综合整个实验结果表明,此方法可以很好的检测饲料中含有的微量木聚糖酶。The purpose of the trial is to determine activities of xylanase in feed by agar diffusion method.The results indicated that the calibration curve showed well linear relations.Meanwhile,there was no significant difference (P 〉 0.05) between test value and theory value in the way to dilute the xylanase by starch,which meaned that the method had high veracity.When the feed was used to dilute the xylanase ,the enzyme activity measured in the trial was a bit less than theory value,however,which has little effect of enzyme activity in the feed.Therefore,the experiment showed that agar diffusion method was of great capability to deterimine trace amount xylanase in feed.
