摘 要:目的对胃癌1号染色体1q43区域的微卫星位点进行杂合缺失(LOH)研究,为筛选此区域内可能存在的胃癌相关抑癌基因提供依据。方法4对荧光标记的微卫星引物(D1S1594、D1S2785、D1S304、D1S321)覆盖1q43区域与96例胃癌患者的肿瘤组织及正常组织进行多重聚合酶链反应(PCR)。产物经毛细管电泳后进行LOH分析。结果该区域所测位点平均杂合缺失率17.9%,其中D1S1594位点最高,杂合缺失率为26.5%;D1S2785位点杂合缺失率最低为7.7%。1q43区域各位点的杂合缺失率与性别、年龄、肿瘤大小及TNM分期无明显相关。结论在1q43区域内发现一个高频LOH区域,即D1S1594及D1S2785位点之间约1cm区域,提示该区域内存在与胃癌相关的抑癌基因。Objective To screen the candidate tumor suppressor genes (TSGs) related to gastric cancer (GC) in 1q43 region. Methods Four fluorescent labeled polymorphie markers were analyzed in the samples of GC and adjacent normal tissues obtained during operation performed on 96 GC patients ,60 males and 36 females. The PCR products were electrophresed for loss of heterozygosity (LOH) scanning and analysis. Results The average LOH frequency in the lq43 region was 17.9% ,with the highest LOH frequency of 26.5% at the D1S1594 locus,and lowest LOH frequency of 7.7% at D1S1594 locus. There was no relationship between LOH frequency and clinicpathologcial data such as the age and sex of the patients,the size and TNM stage of the tumor. Conclusion One high LOH region between D1S1594 and D1S2785 (1q43, about 1 cm) had been detected in GC, suggesting the existence of TSGs related to GC.
