摘 要:对褐牙鲆精子部分生理特性进行了研究,结果表明,褐牙鲆精浆pH值为(7.90±0.26),精子密度为(1.03×1010)/mL,精浆渗透压为(308.50±7.85)m Osm/kg,K+在精浆中的浓度为(27±3)mmol/L,Na+在精浆中的浓度为(71±5)mmol/L。对不同盐度和pH与褐牙鲆精子活力和寿命的关系进行了研究,结果表明,在盐度为35,pH为8.0时,精子的活力最高,分别达到(93.33±5.67)%和(89±3.33)%,与其生活环境相适应;褐牙鲆精子寿命为(23±2)min,与已报道其它海水鱼精子寿命相近。分别采用室温(25℃)和低温(4℃)保存精子,发现精子在室温条件下,可存活4 d,在低温条件下可以存活7 d,证明低温可有效延长精子存活时间。另外,分别采用4种不同稀释液(S1、S2、S3、S4)与4种不同抗冻剂(DMSO、EG、Gly、PG)配制成冷冻保存液对褐牙鲆精子进行超低温冷冻保存研究。结果表明,以S3液作为稀释液,16%的DMSO作为抗冻剂,利用快速降温法对精子进行超低温冷冻保存,38℃水浴快速解冻,解冻后的精子获得最高的活力为(84±3.67)%,此方法可用于褐牙鲆精子的长期保存。Some physiological characteristics of Paralichthys olivaceus sperm were studied. The results showed that the pH of Paralichthys olivaceus sperm was (7.90 ±0.26), density of sperm was ( 1.03 × 10^10)/ml, the osmolality of semen was (308.50 ± 7.85 ) m Osm/kg. The concentration of K^+ in semen was (27 ± 3 ) mmol/ L, concentration of Na ~ was ( 71± 5 ) mmol/L. The relation between salinity ( or pH ) and Paralichthys olivaceus sperm motility was also studied, the results demonstrated that sperm had the best motility of (93.33 ± 5.67) % under salinity of 35 and motility of ( 89 ± 3.33 ) % at pH 8.0, which adapt to the environment. The life-span of Paralichthys olivaceus sperm was (23±2) min, and it was similar to the life-span of other reported marine fishes. The life-span of Paralichthys olivaceus sperm was 4 d under room temperature (25℃ ), whereas it was 7 d under low temperature (4 ℃ ). It shows that low temperature could effectively extend the survival time of sperm. Four different diluters ( S1, S2, S3, S4) and cryoprotectants ( DMSO, EG, Gly, PG) were used for cryopreservation of Paralichthys olivaceus sperm study, results showed thawed sperm (38 ℃ water) had highest motility ( 84 ± 3.67 ) % under the S3 diluter with 16% DMSO as cryoprotectant. This method can be used for long-term preservation of Paralichthys olivaceus sperm.
