桃核承气改良方对兔腹主动脉内膜损伤后血管内皮细胞PDGF-B和STAT3表达的影响

摘　　要：[目的]研究中药桃核承气改良方对家兔血管内皮损伤后PDGF-B和STAT3的影响。[方法]健康新西兰大耳白兔40只，体重2～2.5kg，雌雄不拘。将动物分模型组（n=10）、中药组（n=10）、西药组（n=10）、空白组（n=10），模型组、中药组、西药组高酯饲料喂养3周后行腹主动脉内膜剥脱术造模。术后模型组、中药组、西药组正常饲料喂养，中药组予桃核承气改良方灌胃4周，西药组予舒降之、阿司匹林灌胃4周，模型组、空白组予生理盐水灌胃4周。4周后处死家兔取腹主动脉，用免疫组化法观察各组对PDGF-B表达的影响；采用RT-PCR法测定血管中STAT3水平。[结果]与模型组比较，中药组使血管壁PDGF-B和STAT3表达下降（P〈0.05），中药组与西药组之间无明显差异（P〉0.05）。[结论]桃核承气改良方对PDGF-B和STAT3有拮抗作用。[Objective] Taohe Chengqi Chinese side to improve on the rabbit vascular endothelial injury after PDGF-B and STAT3. [Methods] The health of New Zealand white rabbits 40 and weighing 2 ~ 2.5kg, informal male and female. Animal model of sub-groups （n=10）, the Chinese Medicines Board （n=10）, western medicine group （n=10）, a blank group （n=10）, model group, the Chinese Medicines Board, the group of western high-feeding after 3 weeks Aortic endarterectomy model. After the model group, the Chinese Medicines Board, the normal diet group of western medicine, traditional Chinese medicine to the group to improve the Taohe Chengqi orally for 4 weeks, the group of western medicine to Zocor, aspirin orally for 4 weeks, the model group, to the blank group Oral saline for 4 weeks. 4 weeks after the death rabbit abdominal aorta taken by immunohistochemistry to observe the group of PDGF-B expression, using RT-PCR method blood vessels in the level of STAT3. [Results] Compared with the model group, so that the blood vessel wall Medicines PDGF-B and STAT3 expression decreased （P〈0.05）, traditional Chinese medicine and western medicine group was no significant difference between groups （P〉 0.05）. [Conclusion] Taohe Chengqi improved side PDGF-B and STAT3 there is antagonism.
