摘 要:目的:探讨乙酰肝素酶(HPSE)反义寡核苷酸(AS-ODN)对人鼻咽癌CNE-2细胞HPSE基因表达的影响及其细胞侵袭力的抑制作用。方法:设计合成HPSE AS-ODN转染CNE-2细胞进行培养24h,采用完整细胞原位杂交法检测CNE-2细胞HPSE mRNA表达;同时采用基质凝胶侵袭实验对转染CNE-2细胞的侵袭力变化进行检测。并设正常对照组和脂质体对照组。结果:AS-ODN各组的HPSE mRNA表达水平显著低于对照组,差异有统计学意义(P<0.05)。且AS-ODN各浓度组间的HPSE mRNA表达水平差异亦有统计学意义(P<0.05)。AS-ODN在终浓度分别为0.1μmol/L、0.2μmol/L和0.4μmol/L时对CNE-2细胞侵袭力抑制率分别为56.3%、78.7%和91.2%。结论:HPSE AS-ODN通过下调HPSE基因表达可显著抑制CNE-2细胞侵袭力。Objective:To evaluate the inhibitory effect of heparanase antisense oligodeoxynucleotides (AS -ODN) on gene expression of heparanase and the invasiveness of human nasopharyngeal cancer CNE - 2 cell lines. Methods: One AS - ODN against heparanase mRNA site was transfected into CNE -2 cells, then the expression of heparanase mRNA in CNE - 2 cells was detected by in situ hybridization. Meanwhile Matrigel invasive assay was used to measure the inhibitory effect of heparanase AS - ODN on the invasiveness of CNE - 2 cells. Results: The heparanase mRNA expression and invasiveness of CNE - 2 cells treated with AS - ODN of different concentrations were significantly decreased as the AS -ODN concentration increasing. There was a significantly difference between control group and each group of AS - ODN respectively ( P 〈 0.05 ) , between lipofectamine group and each group of AS - ODN respectively (P 〈 0.05) , and among the groups of AS - ODN ( P 〈 0.05 ) . Besides, the difference of inhibitory effect was significant among the ceils treated with different AS -ODN concentrations (P 〈 0.05). The inhibition rates of CNE - 2 cell invasiveness were 56.3%, 78.7% and 91.2% treated by AS - ODN at final concent -ration of 0.1 μmol/L, 0.2 μmol/L and 0.4 μmol/L respectively. Conclusion: By down - regulating the expression of heparanase gene, heparanase AS - ODN has significantly inhibitory effect on the invasiveness of human nasopharyngeal cancer CNE -2 cell lines .
