宫颈癌和食管癌细胞DNA损伤后相关基因表达的改变

摘　　要：目的:研究宫颈癌和食管鳞状细胞癌(ESCC)细胞DNA损伤前后相关基因的表达。方法:宫颈癌Hela细胞和5种ESCC细胞(YES 2、KYSE 30、YSE 70、KYSE 150和EC 9706)用10 Gy X-ray照射,用顺铂处理ESCC细胞,以达到DNA损伤。提取DNA损伤前后各细胞的RNA和ESCC细胞系的蛋白。用RT-PCR方法检测Aurora-A、p21、Gadd 45a、Bax和Pirh-2基因的转录表达,用Western blot和RT-PCR方法检测ESCC细胞p53基因转录和蛋白表达。结果:Hela细胞经X线照射后Aurora-A、Pirh2基因表达减弱,p21、Gadd45a表达轻度上升;ESCC细胞X线照射后YES 2中Aurora-A表达增强,其余4种细胞Aurora-A表达减弱;顺铂处理后KYSE 30、KYSE 150细胞的p53蛋白表达和p53、p21表达升高,其余基因和另外3种细胞的蛋白及基因表达没有变化。结论:不同种类的肿瘤细胞系和同一肿瘤不同细胞系细胞对DNA损伤有不同反应。OBJECTIVE： To study the gene expressions of Hela cell line and five esophageal squamous cell carcinoma （ESCC） cell lines after DNA damage. METHODS： X-ray （10 Gy） and different concentrations of cisplatin were used to damage DNA of ceils. RNA was isolated from Hela cell line and five ESCC cell lines. The total protein was isolated from the ESCC cell lines. The expressions of Aurora-A, p21, Gadd 45a, Bax and Pirh-2 transcript in these samples were examined with RTPCR. Meanwhile, the expressions of p53 transcript and protein in the five ESCC cell lines were examined with Western blotting and RT-PCR. RESULTS： Aurora A gene was detected in Hela cell line and five ESCC cell lines by RT PCR. After exposed to X ray, Aurora-A gene was found to be up-regulated in the Hela celi line and YES 2 cell line, down-regulated in the other cell lines. In Hela cell line, p21 and Gadd45 genes were found to be up regulated, Pirh-2 was downregulated after X-ray treated. In the five ESCC cell lines, neither p53 protein nor p53 gene was induced in KYSE 70 after exposed to X-ray or Cisplatin. Only bax and p21 genes were found to be up regulated slightly after the cisplatin treatment in KYSE 70. CONCLUSION： Ionizing radiation may induce different genes＇ transcripts in Hela cell line and different ESCC cell lines.
