摘 要:建立高效液相色谱-蒸发光散射(HPLC-ELSD)测定酵母浸膏中游离L-精氨酸(L-Arg)的检测方法。采用Alltima C18(250 mm×4.6 mm i.d.,5μm)色谱柱,以5.0 mmol/L七氟丁酸的0.2%(体积分数)三氟乙酸溶液和乙腈(体积比为93.3∶6.7)为流动相,流速为0.6 mL/min,ELSD漂移管的温度为110℃,载气流速为2.8 L/min。在该色谱条件下,L-精氨酸浓度20.0~500 mg/L时,其峰面积的自然对数与相应的浓度的自然对数线性关系良好(R2=0.9995),回收率97.57%~103.5%,精密度RSD<2.39%。该方法能简便、快速、准确地测定酵母浸膏中L-精氨酸的含量,亦可为其他有机氮源及发酵液中游离L-精氨酸分析提供参考。High performance liquid chromatograpHy (HPLC)coupled with evaporative light scattering detector (ELSD-2000)was used for the determination of L-arginine in yeast extract without derivatization. A AIItima C18 (250 mm ×4.6 mm i.d., 5 μm) was used with the mobile pHase of 0.2% trifluoroacetic acid water solution containing 5.0 mmol/L heptafluorobutyric acid and acetonitrile(the ratio of volume was 93.3 to 6.7). The flow rate was at 0.6 mL/min. The drift tube temperature of ELSD was set at 110 ℃, and with the nitrogen flow rate of 2.8 L/rain. The calibration curves were showed good lineadty(r^2=-0.9995) within the concentration range of 20.0-500 g/L for L-arginine. The average recoveries were ranged from 97.57% to 103.5% and the standard deviation was less than 2.39%. The method was evaluated as rapid, accurate and simple. The validated method is successfully applied to quantify L-arginine in yeast extract which provides a new basis of overall assessment on quality of other organic nitrogen source and determination of L-arginine in fermentation broth.
