摘  要:本文对代谢工程改造红色糖多孢菌ZL1004进行了摇瓶发酵试验,摇瓶发酵结束时重组菌红霉素生物效价与工业生产菌HL3168相比提高20%,红霉素A组分比例由84%提高到94%,红霉素A浓度3.98mg/ml,相比出发菌3.25mg/ml提高22%,重组菌中红霉素B、C与出发菌相比分别减少了51%和58%,实现了红霉素组分的优化。对重组菌摇瓶发酵过程生理代谢进行了初步的研究,表明重组菌生理代谢特性与原出发菌没有明显差异。对重组菌发酵过程中红霉素组分的变化规律进行分析,发现红霉素A、B、C组分的变化规律与eryK和eryG基因转录水平能较好地对应。In this paper, the performance of recombinant industrial Saccharopolyspora erythraea strain ZL1004 was studied in shake flasks. The ratio of Er-A was increased from 84% to 94% with a final concentration of 3.98mg/ml which was 22% higher than that of the original HL3168 strain (3.25mg/ml). Furthermore, the concentrations of Er-B and Er-C were decreased significantly by 51% and 58% respectively. The study on the primary physiological characteristics of recombinant strain ZL1004 showed no apparent difference in glucose, nitrogen, phosphoric consumption rate in comparison with HL3168 strain. The analysis of Er-A, Er-B and Er-C ratios of both ZL1004 and HL3168 in the culture broth during the fermentation process showed a good correlation with the result of transcription level of the eryK and eryG genes in these strains.
